Function

function

Putative OTU-type deubiquitinase (PubMed:26588485).
Catalytically inactive towards all diubiquitin molecules and long K48- and K63- linked ubiquitin chains in vitro (PubMed:26588485).
Potential modulator of apoptosis (PubMed:26588485).

Caution

Both this protein and otu are putative deubiquitinases with homology to other OTU-type deubiquitinases (EC:3.4.19.12), although the nucleophile cysteine residue of its active site has been mutated to serine (Ser-40). Initial characterization showed a lack of deubiquitinase activity towards all diubiquitin molecules and long K48- and K63- linked ubiquitin chains, even when the nucleophile was mutated back to a cysteine (PubMed:26588485).
Subsequent characterization using a more sensitive assay did detect low level deubiquitinase activity for otu (this protein was not retested). Deubiquitinases that are serine proteases instead of cysteine- or metallo- proteases have to date not been described
