Q6LXQ1 · FDHA1_METMP

Function

function

Catalyzes the oxidation of formate to carbon dioxide, with coenzyme F420 as the electron acceptor (PubMed:18791018).
In vitro can also use methyl viologen as electron acceptor (PubMed:18791018).

Miscellaneous

The genome of M.maripaludis harbors two sets of genes encoding the F420-dependent formate dehydrogenase (Fdh), fdhA1-fdhB1 (Fdh1 isozyme) and fdhA2-fdhB2 (Fdh2 isozyme) (PubMed:18791018).
The Fdh1 isozyme is the primary Fdh and the Fdh2 isozyme may not play a major role (PubMed:18791018).

Catalytic activity

Cofactor

Protein has several cofactor binding sites:
[4Fe-4S] cluster (UniProtKB | Rhea| CHEBI:49883 )

Note: Binds 1 [4Fe-4S] cluster.
Mo-bis(molybdopterin guanine dinucleotide) (UniProtKB | Rhea| CHEBI:60539 )

Note: Binds 1 molybdenum-bis(molybdopterin guanine dinucleotide) (Mo-bis-MGD) cofactor per subunit.
Zn2+ (UniProtKB | Rhea| CHEBI:29105 )

Phenotypes & Variants

Disruption phenotype

The deletion mutant cannot grow with formate and produces H2 poorly (PubMed:18791018, PubMed:23335420).
The fdhA1-fdhA2 double mutant is unable to utilize formate for either growth or H2 production (PubMed:18791018).
