Function
function
Catalyzes the oxidation of methanol to formaldehyde in the presence of lanthanides (Ln) (PubMed:30132076).
Contributes to methane/methanol metabolism when La3+ is present in the natural environment of the bacterium, allowing bacterial growth with methane as carbon and energy source. Thereby is an essential enzyme for Ln-dependent methylotrophy (PubMed:26858104).
Uses a specific cytochrome cL, encoded by the adjacent gene in the locus, as electron acceptor (Probable)

Miscellaneous
The methanotroph M.buryatense can produce 2 methanol dehydrogenases (MDH), namely, MxaFI, which is Ca2+-dependent, and XoxF, which is lanthanide-dependent. Lanthanides, abundant in the earth's crust, act as a regulatory switch between the two MDH enzymes, since micromolar amounts of lanthanides are sufficient to suppress MxaFI expression while increasing XoxF expression, even in the presence of excess calcium. The lanthanide concentrations found in the environment are likely sufficient to repress mxa transcription, and XoxF is likely the major functional MDH in M.buryatense in the natural environment.
Catalytic activity
- 2 Fe(III)-[cytochrome cL] + methanol = 2 Fe(II)-[cytochrome cL] + formaldehyde + 2 H+This reaction proceeds in the forward direction.
Cofactor
Protein has several cofactor binding sites:
Note: Binds one lanthanum ion per subunit.
Note: Binds 1 PQQ group per subunit.

Disruption phenotype
Cells lacking this gene display a wild-type growth rate in the absence of lanthanum (when the MxaFI enzyme is maximally expressed) but show a marked growth defect (about a 70% decrease in growth rate) in the presence of lanthanum (a condition under which mxa expression is significantly reduced).
