Function
function
Cytochrome P450 monooxygenase; part of the gene cluster that mediates the biosynthesis of the sesterterpene variecolin (PubMed:38261827).
The first step in the pathway is performed by the variecoladiene synthase vrcA that possesses both prenyl transferase and terpene cyclase activity, converting isopentenyl diphosphate and dimethylallyl diphosphate into geranylfarnesyl pyrophosphate (GFPP) and then converting GFPP into the tetracyclic variecoladiene (PubMed:38261827).
The cytochrome P450 monooxygenase vrcB then catalyzes multiple oxidations at C-5 and C-20 positions to yield variecolin (PubMed:38261827).

Catalytic activity
- 4 O2 + 4 reduced [NADPH--hemoprotein reductase] + variecoladiene = 4 H+ + 6 H2O + 4 oxidized [NADPH--hemoprotein reductase] + variecolinThis reaction proceeds in the forward direction.
Cofactor
Biotechnology
Variecolin and its derivatives produced by employing P450 monooxygenases from other fungal terpenoid pathways exhibit anticancer activity, thus, the skeleton of variecolin could be considered a privileged scaffold for developing anticancer agents.
Pathway
Secondary metabolite biosynthesis; terpenoid biosynthesis.
